Recent studies have highlighted the complexities and challenges in managing myositis. Deveci et al. (2025) reported on two cancer patients who developed myositis with symptoms resembling myasthenia gravis after treatment with pembrolizumab, an immune checkpoint inhibitor. Despite aggressive treatment, both patients succumbed to complications, highlighting the severity of this immune-related adverse event.

In another study, Ikenaga et al. (2025) discovered that in inclusion body myositis (IBM), a loss of TDP-43 splicing repression leads to the production of cryptic peptides, which could serve as potential biomarkers for the disease. This finding suggests a new avenue for therapeutic strategies aimed at restoring TDP-43 function.

Furthermore, Potter et al. (2025) investigated immune-mediated myositis (IMM) following gene therapy with delandistrogene moxeparvovec in patients with Duchenne muscular dystrophy (DMD). They found that T cell responses to specific micro-dystrophin peptides were implicated in IMM, particularly in patients with certain genetic deletions, indicating a need for careful patient selection and monitoring in gene therapy treatments. These studies collectively underscore the importance of early detection and tailored management in myositis to improve patient outcomes.
